Standardized protocol development for integrated construction of Korean Genome and Epidemiology Study

Abstract
Purpose : Korean Genome and Epidemiology Study (KoGES) has evolved with various components which became sometimes inconsistent, and at this stage requires mid-long-term objectives and strategic planning. This study aims at establishing mid-long-term strategy, standardized protocol for integration, and a model for coordinated management.

Methods : Literature review, analysis of existing protocols and tools, international expert consultation meetings, and new tool development were conducted. Working groups developed and proposed guidelines for strategic design, data collection and management protocol, genome analysis and statistical analysis, and compliance to research ethics.

Results : Disease groups with the highest burden include cancer, cardiovascular digestive, diabetes, respiratory, neurophychiatric, and musculoskeletal disorders. For the prevention and control of these diseases, KoGES should generate¡¡knowledge-base for genetic suceptivility and gene-environment interactions.¡¡Comprehensive strategy include
1)individual-family axis, 2)early marker-clinical outcome-sequele axis, 3)growth, development and aging life-course axis. Design priorities are set to large-scale cohort base construction and integration of small cohorts. Genome-wide SNP association study will be the key strategy. Unified questionnaire format with OMR scanning and web-based
participant support system have been developed. Research ethics guidelines are also proposed.

Conclusions : This study provided a strategic design and methods for data collection and analysis. Future KoGES will attain its goals through these integrated management of its diverse components.
